Fragment Screening by Capillary Electrophoresis
Selcia has established a proprietary approach using capillary electrophoresis (CE) to detect the weak binding interactions between fragments and therapeutic targets.
Capillary electrophoresis was pioneered as a sensitive and robust screening technique in the 1990's for high throughput screening (HTS). Selcia has acquired the associated IP and adapted it for fragment screening, creating the CEfrag™ Screen.
CE separates neutral and charged molecules by their mobility in a capillary filled with buffer and subjected to a high-voltage field.
The fragment-target interaction is measured through competitive displacement of a probe ligand by a fragment. Detection occurs by UV or LIF.
CEfrag™ is a particularly powerful tool for identifying hits on challenging biological targets, such as protein-protein interactions.
Fragment hits are identified by monitoring a change in the electrophoretic profile of a probe ligand (PL). For high affinity (low off-rate) PLs, a reduction in a PL:target complex is monitored. For weak affinity (high off-rate) PLs, a PL mobility shift is monitored.
